چکیده

Submitted for the MAR05 Meeting of The American Physical Society Dielectric Properties of Live Yeast Cells Expressed with the Motor Protein Prestin JOHN MILLER, DHARMAKEERTHI NAWARATHNA, DAVID WARMFLASH, University of Houston, FRED PEREIRA, WILLIAM BROWNELL, Baylor College of Medicine — We report on the linear and nonlinear dielectric properties of budding yeast (S. cerevisiae) cells, one strain of which has been genetically modified to express prestin. This motor protein plays a crucial role in the large electromotility exhibited by the outer hair cells (OHCs) of mammalian inner ears. Live cell suspensions exhibit enormous dielectric responses, which can be used to probe metabolic activity, membrane potential, and other properties. We observe a broad peak, centered around 20 kHz, in the normalized difference in dielectric responses between the two strains, and also observe substantial differences in nonlinear harmonic responses. Our data appears to correlate with measurements showing piezoelectric resonances in OHCs. These results suggest that dielectric probes can be used to study the electrical properties of prestin and other proteins, either expressed in live cells or (from our other recent studies on tubulin) as pure protein suspensions. Yeast Two Hybrid cDNA Screening of Arabidopsis thaliana for SETH4 Protein Interaction

SETH4 coding sequence with 2013 bp is a member of gene family expressed in gametophytic tissues of Arabidopsis thaliana. This fragment was PCR amplified using Kod Hi Fi DNA polymerase enzyme. This fragment was cloned into pGBKT7 bate vector and transformed E. coli DH5? cells containing vector were selected on LB medium containing Kanamycin. Finally, pGBKT7-SETH4 bate was transformed into yeast ...

Expression of the VP2 gene of classical D78 infectious bursal disease virus in the methylotrophic yeast Pichia pastoris as a secretory protein

Infectious bursal disease virus (IBDV) is the causative agent of Gumboro disease, an infectious disease of global economic importance in poultry. The expression of heterologous proteins in P.pastoris is fast, simple and inexpensive. In this study, VP2 encoding gene of classical D78 IBDV was amplified using reverse transcription (RT) polymerase chain reaction (PCR) and cloned into pPICZαA vector...

The effect of titanium dioxide and molybdenum oxide on the properties of sodium potassium niobate piezoceramic

Sodium potassium niobate piezoceramics are considered as lead-free piezoelectric due to their good properties. In this study, the effect of titanium dioxide and molybdenum oxide as dopant on the densification behavior, piezoelectric and dielectric properties of lithium doped sodium potassium niobate ((K0.5Na0.5)0.94Li0.06NbO3, KNNL) was investigated. For this purpose, (K0.5Na0.5)0.94Li0.06Nb0.9...

Effect of Different Levels of Live Yeast in a High Concentrate Diet on Performance, Blood Constituents and Immune System Status of Zandi Lambs

A feeding trial with twenty-seven male weaned Zandi lambs (initial body weight 27.1±0.38 kg) was conducted to evaluate the effect of different levels of Saccharomyces cerevisiae SC47 in diet containing high concentrate (85%) on the growth performance, blood parameters and immune system status. Lambs were allocated to one of three treatment diets in a completely randomized design with 3 replicat...

A Combination Method of Centrality Measures and Biological Properties to Improve Detection of Protein Complexes in Weighted PPI Networks

Introduction: In protein-protein interaction networks (PPINs), a complex is a group of proteins that allows a biological process to take place. The correct identification of complexes can help better understanding of the function of cells used for therapeutic purposes, such as drug discoveries. One of the common methods for identifying complexes in the PPINs is clustering, but this study aimed ...
